Scientific Background

Semax is a synthetic heptapeptide analog of ACTH(4–10) — the core active fragment of adrenocorticotropic hormone responsible for neurotrophic signalling — extended with a Pro-Gly-Pro C-terminal sequence that protects against enzymatic degradation. Developed at the Institute of Molecular Genetics of the Russian Academy of Sciences, Semax has been in clinical use in Russia and Ukraine for over two decades as a neuroprotective and cognitive-enhancing agent.

Semax's primary mechanism centres on rapid upregulation of BDNF (Brain-Derived Neurotrophic Factor) and NGF (Nerve Growth Factor) in the hippocampus and frontal cortex — key regions for learning, memory, and executive function. It also enhances dopaminergic and serotonergic neurotransmission, improves cerebral blood flow, and activates neuroprotective gene expression within hours of administration.

In clinical research, Semax has demonstrated accelerated functional recovery in ischaemic stroke patients, reduced cognitive deficits in hypoxia models, and improved attention and memory in healthy subjects. Compared to Selank (primarily anxiolytic/mood-stabilising) and Adamax (stronger but longer-acting BDNF inducer), Semax occupies the cognitive enhancement and neuroprotection space with a well-characterised safety profile from decades of Russian clinical use.

Intended Research Use

  • BDNF and NGF upregulation for neuroplasticity research
  • Neuroprotection in ischaemic stroke and hypoxia models
  • Cognitive enhancement — attention, memory, and learning studies
  • Dopaminergic and serotonergic neurotransmission modulation
  • Cerebral blood flow improvement research
  • ACTH(4–10) analog pharmacology
  • Comparative nootropic peptide research (Semax vs. Selank vs. Adamax)
