SNAP-8 is a synthetic octapeptide designed as an extended and enhanced analog of the hexapeptide Argireline (Acetyl Hexapeptide-3). It replicates the N-terminal sequence of the synaptosomal-associated protein SNAP-25 — a critical component of the SNARE complex responsible for vesicular neurotransmitter release at neuromuscular junctions.
By competing with SNAP-25 for incorporation into the SNARE complex, SNAP-8 reduces the assembly efficiency of the synaptic fusion machinery, thereby decreasing acetylcholine release at neuromuscular junctions. This results in a partial, reversible reduction in muscle contractility — analogous to the mechanism of botulinum toxin (Botox), but acting at the SNARE protein level rather than through proteolytic cleavage. The effect is therefore significantly milder and fully reversible, producing muscle relaxation without immobilisation.
